Preparation and Application of Nucleic Acids and Their Compositions with Hair Loss Prevention and Sebum-Control Functions
Abstract:
The present invention relates to the preparation and use of nucleic acids and compositions thereof having anti-hair loss and sebum-control effects, and belongs to the technical field of daily chemical products. The small interfering RNA sequences designed in the present invention exhibit significant silencing effects on the AR target gene. The compositions prepared by using ionic liquids or ionic liquids in combination with lipids provide effective delivery of water-soluble nucleic acid molecules, and can penetrate cells and enter the nucleus to achieve AR silencing. The compositions also exert significant regulatory effects on downstream target genes or proteins regulated by AR, thereby providing more comprehensive anti-hair loss and sebum-control effects. In addition, the compositions of the present invention exhibit good transdermal permeation-enhancing properties, enabling penetration through the superficial skin layers to the dermis, and thus showing significant anti-hair loss and sebum-control efficacy, as well as good antibacterial activity against Propionibacterium acnes. The compositions of the present invention are nucleic acid materials with triple functions of anti-hair loss, sebum control, and anti-acne, thereby expanding the applications of AR-targeting nucleic acids.
Efficacy
Clinically Proven Anti-Hair-Loss Efficacy
In a 32-participant trial, UNISOUL® RNAiG visibly reduced hair loss within 7 days, with continued improvement through Days 14 and 28.
Day 7: Hair fall reduced by 27.9% from baseline.
Day 14: Hair fall reduced by 36.7% from baseline.
Day 28: Hair fall reduced by 48.6% from baseline.

UNISOUL® RNAiG effectively targets the root causes of hair loss by silencing androgen- and inflammation-related pathways. In DHT-induced cell studies, it significantly reduced AR, DKK-1, TGF-β2, BMP4, IL-1α, IL-8, and TNF-α expression while boosting β-catenin, key markers of follicle activation. The result: visibly stronger, thicker, and healthier hair from the root.

UNISOUL® RNAiG delivers asiRNA with exceptional precision—achieving over 70% reduction in AR expression (p<0.001).
Proven to outperform free asiRNA, it ensures deeper delivery, stronger efficacy, and visible cellular results.
Fluorescence microscopy revealed that asiRNA encapsulated in UNISOUL® RNAiG achieved 5–12× higher fluorescence intensity compared with the nucleic acid PBS solution, demonstrating significantly enhanced transdermal penetration and intracellular delivery efficiency.
