Invention Name:
A preparation method of a siRNA supramolecular microcapsule with whitening effect and its application
Efficacy
UNISOUL® RNAiW targets and silences the expression of MITF, a master regulator of melanogenesis, to effectively address various types of hyperpigmentation, including chloasma.
Compared to traditional whitening ingredients, UNISOUL® RNAiW demonstrates superior melanin-inhibiting efficacy. Clinical data show noticeable improvements in the number and size of brown and UV-induced spots after just 14 and 28 days of use.
When compared to the benchmark whitening agent kojic acid, treatment with 0.8% UNISOUL® RNAiW resulted in a 64.7% greater reduction in melanin content. Formulated at 5% concentration in the UNILIPO essence, the brightening effect is both rapid and visible.
After 28 days of use, UNISOUL® RNAiW was shown to reduce melanin by 2.44% and increase overall skin brightness by 1.53%, confirming its role as a highly effective active for pigmentation control and skin tone improvement.
Advantages
The application of supramolecular technology significantly enhances the transdermal absorption efficiency of siRNA, enabling deeper skin penetration and improved bioavailability for optimal performance.

A 12-week clinical study demonstrated that UNISOUL® RNAiW achieved superior melasma reduction compared to 10% magnesium ascorbyl phosphate (MAP) by effectively inhibiting the expression of MITF, a key regulator of melanogenesis.
We developed supramolecular liposomes for UNISOUL® RNAiW, which demonstrate enhanced performance compared to conventional liposome encapsulation. This advanced delivery system significantly improves the transdermal absorption efficiency of siRNA, ensuring more effective penetration and bioactivity.

UNISOUL® RNAiW significantly inhibited the four most critical genes, MITF, TYR, TRP-1, and TRP-2, at both the mRNA transcription level and the protein expression level.

Right image: The silencing effect of siRNA on multiple key genes involved in the melanin synthesis pathway. In the gel electrophoresis results, thicker and darker bands indicate higher gene expression, while thinner and lighter bands indicate reduced expression, confirming the effective downregulation of melanin-related genes.

At a concentration of just 0.8%, UNISOUL® RNAiW reduced melanin content by 64.7% more than kojic acid, a gold-standard whitening agent. Notably, our pterostilbene essence is formulated at 5%, offering even greater potential for visible brightening and pigment correction.

Statistical data indicate that just 28 days of treatment with UNISOUL® RNAiW significantly reduced skin melanin levels by 2.44% and enhanced skin brightness by 1.53%, demonstrating its efficacy in improving overall skin tone and clarity.

After 14 and 28 days of treatment with 5% UNISOUL® RNAiW, there was a visible reduction in both the number and size of brown spots and UV-induced pigmentation, demonstrating its effective skin-brightening performance.
